  2. Watch 1 peer video, and create a new document with peer feedback: share that with me and the other person. I will be looking for this today.
  3. Please upload your script for your Fish DNA Barcoding Lab to your Google Doc shared folder with me, OR upload your preliminary lab write-up for that lab in the foldershared with me.
  4. Upload your PCR Script or prelim rough draft video to the shared folder
  5. Upload the plasmid script and/or your prelim rough draft video to the shared folder

Bold Systems Update: 
I did hear back from Alexandra at BOLD - she can only delete all traces for a sample, not selectively delete. I attempted to upload Amber and Zada's samples, and Lucie and Addy's as well. We can check on how to do that. She said she will call us, but I don't know when - it is her day off and I am not sure what time zone she is in.....

Review the Ligation Experiment  Protocol, this is a go for Thursday......!!
Next videos, let's figure out a timeline and take a look at where we are at on these.
If time, we will write/peer review scripts.....

Wow, do we have a lot to cover this week! And next....
  • In the interim until Digication is up and running, you all have a folder I created in google docs, let's look at this together quickly, but I know it works because Izzy helped me test it (Thank You Izzy!)
  • So, all videos, lab write-ups, etc, can go into your individual folders
  •  I would like to invite you to share your first videos in the name of learning in our community folder           everyone has access to. Please put pre/post lab questions in our personal folders, BUT PRINT OUT A         HARD COPY FOR THE OLD LADY, (much obliged).
  • Your videos are graded, I have some feedback for you on those, they will be entered into Q this afternoon (at the latest)
  • Tips on work management - set yourself an intermediary timeline, just do it
  • ​Please see the Enrichment tab (built only in my mind at this point, but have faith) for opportunities to  stretch yourself and demonstrate excellence, by the end of the week
  • Next Quiz study page has been created and I am adding thoughts to that on what you will need to know Let me know if you have thoughts, questions, comments as we go along
  • ​Primer Discussion on the reading, let's pick it up where Michael left off on the proteins, fascinating!
  • We will have a lot of content to cover for the Ligation and Transformation components of our GAPDH series. Here is a tentative timeline:
  • PCR Kleen - was going to be today, but due to the college fair it will be tomorrow! Clean the extra primers, Taq, dNTP's, out of our samples
  • Tomorrow: Revisit Ligation, selective media
  • Wednesday: make ampicillin plates (Izzy beware, PPE all around)
  • Thursday: Lecture on restriction enzymes/digest

Fishbarcoding samples have been received by Sequetech, now we wait for results.
Video Skills Log - Amazing. Powerful. Impressive.  I am going through these right now and I am wondering if we might have a few brave souls who would share theirs and allow a class critique? I think this would be very valuable...... At least, please do view two peers videos each, provide a written critique for each person to go into digication along with their video (and my feedback). Let's discuss this.

Let's go over some questions - wobble versus degenerate primers, the same thing different name? What do you think?

What are we doing? Let's take a step back and take a bird's eye view before we move forward. Questions/discussion.

Last 30 min. of the period - I will be in the SS Mike with some of you for Digication training; you will start either 1) your next video skills log or 2) your content log on the lab you didn't write-up or 3) you will work on your formal lab write-up. You must be actively working on one of these things. Please share your work with Andrew and Jiren as they come around to see how you are learning/growing.
