Transformation Lab Today
In down time, peer review PCR Scripts
Ligation component of our lab today, make sure your samples are in the freezer for Monday's transformation lab on Monday.
Bold Systems Update:
I did hear back from Alexandra at BOLD - she can only delete all traces for a sample, not selectively delete. I attempted to upload Amber and Zada's samples, and Lucie and Addy's as well. We can check on how to do that. She said she will call us, but I don't know when - it is her day off and I am not sure what time zone she is in.....
Review the Ligation Experiment Protocol, this is a go for Thursday......!!
Next videos, let's figure out a timeline and take a look at where we are at on these.
If time, we will write/peer review scripts.....
Okay, back on-line..... recovering from all that happened last week!
Our new reagents have been ordered, and will be here Wednesday.
Today, Sequetech and BOLD Database work - we have it figured out!
Discuss grades and videos - feedback?
Look at our enormous task of analyzing our fish barcoding sequences (how do you eat an elephant? One bite at a time!)
Look at our plates...we have growth.
Re-do any assingments, or start

Review our ligation lab protocol, we will be starting on this either Monday or Tuesday.
On Tap: Fish Barcoding.......
1. Discuss how to pour plates, each group will pour 6 plates
Store upside down on the counter
2. If time, polish your assignments due Thursday,
3. Please make sure you put these in you Google Folder that is shared with me
I will pour your LB/amp/IPTG plates for you Friday

Wow, do we have a lot to cover this week! And next....
Check your three-ring binders
Digication Update! Progress, yet not ultimate success, stay tuned. In the meantime,
All BT students start a folder in Google Docs and put you required work in there so that when
Digication is ready, you are ready (this is what we agreed on as a staff in our staff meeting)
Take Stock of where you are at regarding your assignments due next:
1. Content Video Log 1
2. Your first formal lab write-up, please see the tab on lab write-up requirements
3. Your next set of Skills videos
Work Time - 30 minutes
Review the article I gave you yesterday
Read the overview for your Ligation and transformation,
Fishbarcoding samples have been received by Sequetech, now we wait for results.
Video Skills Log - Amazing. Powerful. Impressive. I am going through these right now and I am wondering if we might have a few brave souls who would share theirs and allow a class critique? I think this would be very valuable...... At least, please do view two peers videos each, provide a written critique for each person to go into digication along with their video (and my feedback). Let's discuss this.
Let's go over some questions - wobble versus degenerate primers, the same thing different name? What do you think?
What are we doing? Let's take a step back and take a bird's eye view before we move forward. Questions/discussion.
Last 30 min. of the period - I will be in the SS Mike with some of you for Digication training; you will start either 1) your next video skills log or 2) your content log on the lab you didn't write-up or 3) you will work on your formal lab write-up. You must be actively working on one of these things. Please share your work with Andrew and Jiren as they come around to see how you are learning/growing.
